Senior Scientist, Multi-Specifics, Biologics Engineering

We are seeking a skilled and highly motivated Senior Scientist to enhance our Biological Expression Team. The successful candidate will play a critical role in a dynamic group responsible for performing expression and purification of a wide variety of antibodies, multi-specifics, and complex modalities. The successful candidate will be a key member of our growing team, providing high quality, well-characterized proteins to support biologics discovery across multiple therapeutic indications. This individual will be a well-organized and a highly productive scientist that can design and execute experiments efficiently, analyze, interpret, and present data to multidisciplinary project teams. Collaborate and work with a diverse group of scientists across all therapeutic areas to ensure timely progress in research projects.

BUSINESS AREA

Biologics Engineering is large global organization with team in Gaithersburg MD, and Cambridge, UK responsible for the discovery and optimization of next generation biologics to create biological drug candidates for all key therapeutic areas such as oncology, respiratory and inflammation, cardiovascular, renal and metabolic diseases. Our Waltham team is a new team we are building from the ground up to support both our global responsibilities as well as building an innovation center where we can push the boundaries of sciences and creativity. We're constantly pushing the boundaries of science to deliver life-changing medicines to patients, with a passion for the development of in-house biologics discovery platforms and novel drug modalities to address unmet medical needs.

The Waltham site offers a variety of amenities to help boost productivity and help keep our employees stay happy and healthy. This is where you’ll find newly-designed, activity-based work spaces to suit a variety of working styles while increasing collaboration between teams.

What you'll do:

  • Perform expression and purification of mono- and multi-specific antibodies and complex modalities fusion proteins from sub-milligram to gram scale to support antibody discovery, characterization, and pre-clinical validation of leads.

  • Perform biologics quality assessment utilizing a variety of techniques, such as CE-SDS, SEC-HPLC, and endotoxin assay.

  • Develop and implement purification procedures to improve existing processes and workflows.

  • Maintain and troubleshoot relevant laboratory instrumentations, such as AktaPure, AktaAvant, AmMag.

  • Maintain complete and updated records in electronic lab notebook.

Basic Qualifications:

  • PhD with 2 plus years of experience

  • Or MS Degree with 5 plus years of experience, or BS Degree with 8 plus years of experience in Biologics production.

  • Extensive with transient mammalian expression (HEK293 and CHO) and purification of mono- and bi-specific antibodies, Fab fragments, scFv, VHH, and various antibody-fusion formats. 

  • Extensive hands-on experience with chromatography instrumentation (AktaPure, AktaAvant, AmMag) including ability to develop custom methods and troubleshoot. Experience with automation is preferred.

  • Strong protein characterization and QC skills including analytical SEC, CE-SDS, endotoxin assays.

  • Familiarity with molecular biology techniques, including primer design, gene assembly, Gibson and Golden gate cloning, vector assembly, transfection, and software for sequence analysis.

  • Experience interacting with CMC groups and understanding development upstream and downstream processes, including ultrafiltration, viral clearance, etc. is highly desired.

  • Proven ability to work independently, effectively plan and organize work activities, and prioritize task completion to meet project deadlines in a fast-paced industrial environment.

  • Strong interpersonal and communication skills, ability to collaborate and communicate across disciplines.

Desired Skills

  • Proficiency with various modes of chromatography, including affinity, IEX, mixed mode, HIC, and SEC.

  • Experience in various imaging/multicolor flow cytometry-based instruments and data analysis programs.

  • Experience in tissue culture techniques and functional cell-based assay development.
